We hope to understand how chromatin is organized in a cell and how this structural architecture is coupled to the functional regulation of gene expression. Our primary focus is gene insulation, the phenomenon by which promoters in a given locus are shielded from the effects of all regulatory sequences in a cell except for those within the locus itself, thus resulting in an autonomously functioning genetic element. We attempt to combine structural methods (EM, x-ray, mass spec) with functional experiments (cell-based screens and assays) to develop macromolecular movies of how chromatin controls transcriptional organization.
